1. Differential centrifugation. A method of stepwise centrifugation of particles having different sedimentation rates using different centrifugal speeds and centrifugation times is called differential centrifugation. During the operation, the suspension containing two different particles will be centrifuged at a constant speed to sink the large particles, the supernatant is poured into another centrifuge tube, and the centrifugal force is increased, and the centrifugation is performed for a certain period of time, and the separation is small. The granules are repeatedly separated several times to achieve the purpose of separation. Differential centrifugation is mainly used to separate particles with large differences in size and density.

2. Density gradient centrifugation. The method is further divided into a velocity zone centrifugation method and an equal density zone centrifugation method. The sample is subjected to centrifugal sedimentation or sedimentation equilibrium in a certain inert gradient medium, and the particles are distributed to certain specific positions in the gradient liquid under a certain centrifugal force to form a separation method of different zones. The material to be centrifuged is separated according to its sedimentation coefficient, and the same kind of substance is separated because the molecular deposition rate is faster than that of the small molecule. The density gradient can be prepared by using a ladder mixer. Different concentrations of sucrose solution can be carefully added to the centrifuge tube layer by layer. The higher the concentration is, the higher the concentration is, and the step gradient is formed. During density gradient centrifugation, the position and width of the zone vary with the time of centrifugation. As the centrifugation time increases, the zone becomes wider and wider due to particle diffusion.

The method has the advantages of good resolution, good separation effect and relatively pure particles at one time; wide application range, which can separate particles with poor sedimentation coefficient and particles with certain buoyancy density; The backlog deforms to maintain particle activity and prevent mixing of the formed zones due to convection.

3. Analytical ultracentrifugation

The analytical ultracentrifuge consists mainly of an elliptical rotor, a vacuum system and an optical system. The rotor is connected by a flexible shaft into a high speed drive that allows the rotor to form its own shaft as it rotates. The rotor rotates in a chilled vacuum chamber that houses two chambers: an analysis chamber and a ration chamber. The analysis chamber has a capacity of generally 1 mL and is arranged in a fan shape in the rotor. The working principle is the same as that of a normal horizontal rotor. The analysis chamber has two upper and lower flat quartz windows. The optical system installed in the centrifuge ensures that the sediments in the chamber can be observed during the entire centrifugation.
